Gonadorelin Description1-5

Gonadorelin is a synthetic decapeptide that mirrors the amino acid sequence of naturally occurring gonadotropin-releasing hormone (GnRH). Both substances consist of 10 amino acids arranged in the sequence: pyro-Glu-His-Trp-Ser-Tyr-Gly-Leu-Arg-Pro-Gly-NH₂.

In males, FSH and LH are involved in supporting testosterone production and sperm development. In females, these hormones play a role in managing the menstrual cycle and ovulation. The half-life of gonadorelin ranges from approximately 2 to 40 minutes.

Anastrozole Description6-8

Anastrozole is a selective, non-steroidal aromatase inhibitor. It is absorbed quickly, typically within about 2 hours. Its half-life is around 50 hours, and with daily use it generally reaches steady-state levels in about one week. Anastrozole works by reducing estrogen synthesis and influencing testosterone levels.

Gonadorelin1-5

GnRH is made in the hypothalamus and functions to stimulate the release of follicle stimulating hormone (FSH) and luteinizing hormone (LH) from the anterior pituitary. This increase in FSH and LH may stimulate the production and release of testosterone by the male testes and estrogen by the female ovaries and placenta.

Anastrozole6-8

In postmenopausal women, estrogens are mainly derived from the action of the aromatase enzyme, which converts adrenal androgens (primarily androstenedione and testosterone) to estrone and estradiol. Suppression of estrogen biosynthesis in peripheral tissues and in cancer cells can be achieved by specifically inhibiting the aromatase enzyme. In males, inhibition of the aromatase enzyme results in decreased systemic estrogen levels and increased systemic testosterone levels.

  • Headache
  • Nausea
  • Abdominal discomfort
  • Flushing
  • Dizziness or lightheadedness
  • Injection site reactions such as redness, pain, or swelling

Common anastrozole adverse reactions6,8

  • Acne
  • Hypertension
  • Gastrointestinal (nausea, vomiting, abdominal pain)
  • Headache
  • Weight gain
  • Osteopenia
  • Insomnia

Contraindications1,2,6,8

  • Intolerance or hypersensitivity to gonadorelin or anastrozole
  • Use in pregnant or breastfeeding women is contraindicated

Precautions1,2,6,8

  • Rare instances of hypersensitivity reaction (bronchospasm, tachycardia, flushing, urticaria, induration at injection site) and anaphylactic reactions have been reported following multiple-dose administration.
  • There has been a report of pituitary apoplexy and sudden blindness following gonadotropin-releasing hormone administration to a patient with a gonadotropin-secreting adenoma.
  • Caution in patients with osteoporosis and coronary artery disease
  • Store at controlled room temperature (20°C to 25°C / 68°F to 77°F)
  • Protect from moisture and light
  • Keep in a tightly closed container
