Tri-Test Description1-10

Cypionate 50% / Enanthate 37.5% / Propionate 12.5% Injection is a compounded injectable blend of testosterone esters containing 50% testosterone cypionate, 37.5% testosterone enanthate, and 12.5% testosterone propionate. This ester combination has been used in testosterone replacement therapy (TRT) for the management of male hypogonadism. The formulation is designed to incorporate the differing pharmacokinetic profiles of the three esters to achieve a more consistent and prolonged release of testosterone, which may allow for fewer injections and reduced variability in hormone levels. Testosterone enanthate has an approximate half-life of 7 days, slightly shorter than testosterone cypionate at about 8 days, while testosterone propionate has a half-life of approximately 4.5 days. In contrast, non-esterified testosterone has a half-life as short as 10 minutes, indicating that esterification substantially prolongs testosterone’s half-life.

Mechanism of Action1-10

Testosterone cypionate, testosterone enanthate, and testosterone propionate are esterified forms of testosterone. The esterification occurs on the 17b carbon of the testosterone structure. The esters extend the duration of action and are useful for less frequent, intramuscular, or subcutaneous injections.  Once administered, the esters are cleaved, releasing free testosterone into the bloodstream. Testosterone then binds to androgen receptors in various tissues, exerting anabolic and androgenic effects. Testosterone has a role in influencing the development and maintenance of male secondary sexual characteristics, stimulation of muscle and bone growth, and regulation of libido and mood.

Contraindications1  

  • Known hypersensitivity to testosterone or the respective ester component
  • Known hypersensitivity to grapeseed oil
  • Males with carcinoma of the breast or known or suspected carcinoma of the prostate
  • Pregnant or breastfeeding women
  • Patients with serious cardiac, hepatic, or renal disease

Precautions1  

  • Monitor hematocrit and hemoglobin levels.
  • Regularly assess liver function tests.
  • Evaluate lipid profiles periodically.
  • Monitor for signs of sleep apnea.
  • Use cautiously in patients with a history of cardiovascular disease.

Store at 20°C to 25°C (68°F to 77°F) in a dry place, away from heat and light. DO NOT REFRIGERATE. Do not freeze. Keep out of reach of children.
