Niacinamide / Panthenol / Estriol / Tretinoin – Description1-9

This topical gel combines four active ingredients: niacinamide, panthenol, estriol, and tretinoin—to address multiple features associated with skin aging, photodamage, and barrier dysfunction. It is used primarily in cosmetic dermatology to support improvements in skin texture, tone, elasticity, and hydration, while facilitating cell turnover and collagen remodeling.

Niacinamide1-3 (Vitamin B3) A water-soluble vitamin that may strengthen the skin barrier, reduce inflammation, and improve uneven pigmentation and fine lines supporting healthy skin.

Panthenol4-5 (Precursor to vitamin B5) A humectant and emollient with potential for anti-inflammatory and skin-soothing properties; may support wound healing and hydration.

Estriol6-7 A weak estrogen with potential for skin-rejuvenating properties; used in postmenopausal women or estrogen-deficient states to restore dermal thickness and elasticity.

Tretinoin8-9 Vitamin A derivative and potent retinoid that stimulates skin cell turnover, may help smooth wrinkles, and may help to treat hyperpigmentation and acne.

Niacinamide1-3

  • Inhibits melanosome transfer which may assist in pigmentation
  • Stimulates ceramide production which may strengthen the skin barrier
  • Reduces local inflammation which may calm erythema and acne
  • May enhance dermal fibroblast activity which may promote elasticity

Panthenol 4,5

  • Converted to pantothenic acid (Vitamin B5) in the skin
  • Pantothenic acid can improve skin hydration via humectant properties
  • Promotes fibroblast proliferation and keratinocyte differentiation
  • Anti-inflammatory effects aid in healing irritated or sensitive skin

Estriol 6,7

  • Binds estrogen receptors in dermis and epidermis potentially leading to:
  • Increases in collagen types I and III
  • Enhanced vascularity and hydration
  • Thickening of dermal-epidermal junction, potentially improving firmness

Tretinoin8,9

  • Binds retinoic acid receptors (RARs) which modulate gene expression that may influence directly or indirectly:
  • Accelerated keratinocyte turnover
  • Stimulation of fibroblasts to produce collagen and elastin

Niacinamide 1-3

Mild flushing, tingling, rare allergic contact dermatitis

Panthenol 4,5

Occasional mild irritation in sensitive individuals

Estriol 6,7

Local irritation, breast tenderness, spotting (rare); systemic absorption is low but possible; Cumulative estrogenic side effects are rare but possible with long-term estriol use

Tretinoin 8,9

Peeling, dryness, redness, burning, initial acne flare; photosensitivity is common

Niacinamide 1-3

Contraindication: Hypersensitivity – Precaution: Use cautiously in rosacea or sensitive skin

Panthenol 4,5

Contraindication: Allergy to panthenol – Precaution: Rare allergic reactions

Estriol 6,7

Contraindication: Estrogen-dependent cancers, thromboembolic history, unexplained bleeding – Precaution: Avoid in pregnancy/lactation; use under medical supervision

Tretinoin 8,9

Contraindication: Pregnancy, hypersensitivity, irritated or sunburned skin – Precaution: Use sunscreen; discontinue with severe irritation

Store at 20°C to 25°C (68°F to 77°F) in a dry place, away from heat and light. Keep out of reach of children.
