Ondansetron Description1,2

Ondansetron is a 5-HT₃ receptor antagonist that is used clinically as an antiemetic. It is utilized to prevent and manage nausea and vomiting, particularly in patients undergoing chemotherapy, receiving radiation therapy, or following surgery or other medical procedures.

Mechanism of Action1,2

Serotonin (5-HT₃) receptors are found in the gastrointestinal tract and centrally in the chemoreceptor trigger zone (CTZ) in the brain. Ondansetron prevents serotonin from binding to serotonin receptors in the gastrointestinal tract and the chemoreceptor trigger zone. By preventing serotonin from binding to these receptors, it inhibits vomiting reflexes, specifically when chemotherapy, radiation, and anesthesia is administered.

Contraindications1,2

  • Known hypersensitivity to ondansetron or other 5-HT₃ antagonists.

Precautions1,2

  • QT interval prolongation: avoid in patients with risk factors for QT prolongation including congenital long QT syndrome or electrolyte imbalances. (hypokalemia, hypomagnesemia)—QT interval prolongation can lead to torsades de pointes.
  • Hepatic impairment: In severe hepatic dysfunction, do not exceed 8 mg total daily dose.
  • Store at controlled room temperature (20°C to 25°C / 68°F to 77°F)
  • Protect from moisture and light
  • Keep in a tightly closed container
