Letrozole Tablets Description1,2

Letrozole is a nonsteroidal aromatase inhibitor that works by suppressing estrogen production through inhibition of the aromatase enzyme. It is primarily used in postmenopausal women for the treatment of hormone receptor–positive breast cancer, including both early-stage disease and advanced or metastatic breast cancer.

Mechanism of Action1,2

Letrozole binds to heme group of the cytochrome P450 subunit of the aromatase (enzyme) resulting in a reduction of estrogen (estrone, estradiol and estrone sulfate) biosynthesis in all tissues. The drug decreases estrogen levels by preventing the conversion of androgens to estrogen.

  • Known hypersensitivity to letrozole or any component of the formulation
  • Letrozole is contraindicated during pregnancy

Precautions1,2

  • Reduction in bone mineral density
  • Use letrozole with caution in patients with pre-existing hypercholesterolemia
  • Store at controlled room temperature (20°C to 25°C / 68°F to 77°F)
  • Protect from moisture and light
  • Keep in a tightly closed container
