N-Diindolylmethane (DIM) is a naturally occurring compound produced from the digestion of indole-3-carbinol (I3C), which is present in cruciferous vegetables such as broccoli, Brussels sprouts, and kale. DIM is commonly used as a dietary supplement or compounded into oral capsules or topical formulations for its potential roles in hormonal balance, anti-cancer, and anti-inflammatory support.

Mechanism of Action1-6

DIM may influence estrogen metabolism by shifting the breakdown of estradiol (E2) toward 2-hydroxyestrone (2-OHE1) instead of 16α-hydroxyestrone, a potentially pro-carcinogenic metabolite. carcinogenic metabolite. Scientific studies continue to investigate how this compound interacts with biological pathways, including: 

  • DIM may act as a selective androgen receptor modulator (SARM), helping reduce androgen-driven conditions like acne and hormonal hair loss in some individuals.
  • Aromatase Inhibition (mild)
  • Some studies suggest DIM can mildly inhibit aromatase, the enzyme that converts testosterone to estradiol, making it of interest in estrogen-dominant or androgen-sensitive conditions.
  • Shown to have Anti-Inflammatory and Antioxidant Activity
  • May modulate NF-κB signaling, reducing inflammatory cytokine release. Has exhibited antioxidant properties, scavenging reactive oxygen species (ROS).
  • Shown to promote apoptosis (programmed cell death) and inhibit angiogenesis in certain cancer cells (in vitro)

  • Hormone-sensitive conditions (e.g., breast cancer, endometriosis, fibroids): DIM may alter estrogen activity; consult healthcare provider before use.
  • Anticoagulant use: DIM may have mild antiplatelet effects; monitor bleeding risk.
  • Concurrent hormone therapy: DIM may alter efficacy of oral contraceptives or hormone replacement therapy (HRT).
  • May interact with cytochrome P450 enzymes, affecting metabolism of other drugs.
  • Store at controlled room temperature (20°C to 25°C / 68°F to 77°F)
